When Iago and Othello are by themselves, Iago started to say that Desdemona was having an affair with Cassio save there was little assure that they are together. This is where the problem is rising up. Iagos plan was to cultivate Othello doubt and jealous about Desdemona. What Iago reported to Othello injected into him like a virus. He might kill her for cheating.
As soon as Iago took off, Othello asked himself wherefore he married. He said Iago knows more than himself. What he said was Why did I married? Im sure this good and honest man sees and knows more, much more, than hes telling me. (III, iii, 242-243). Othello fell into Iagos trap and he did it without thinking.

Emilia doesnt know why Iago needed it so badly but Iago told her to give it to him. Iago is spill to plant it in Cassios bedroom so that he could prove to Othello that Desdemona had cheated him.
